Here you can find a basic overview of the parts we used in our construct and our various testing plasmids.

In order to test each of the four modules of our construct, we designed specific testing plasmids that allow us to better observe and characterize the function of our parts. We used Golden Gate assembly to build the plasmids, therefore we have various Level 1 and Level 2 plasmids. There is a description and an image of all composite parts in our testing plasmids.

1. Sensing module:

The table below contains all basic parts and testing plasmids we constructed for the sensing component of our circuit. It is composed of a protein that forms a complex with the sugar xylose and binds a specific promoter.

2. Biofilm overexpression:

Our biofilm overexpression module consists of the following basic and composite parts. On one hand we boost biofilm by inducing the expression of a diguanylate cyclase, on the other hand we knock down a phosphodiesterase using the CRISPRi system.

3. Feedback loop:

We designed a feedback loop which consists of the T7-like RNA polymerase of the Pseudomonas phage Pf-10 and the respective T7-like promoter it binds to. Besides further boosting the biofilm overexpression, the feedback loop also links the separate components of our construct.

4. Kill switch:

Our kill switch consists of a DNA gyrase inhibiting toxin and its respective antitoxin. The toxin is expressed after the T7-like promoter, while the antitoxin - after the xylose promoter.

The transcriptional unit expresses a toxin which is induced by a rhamnose promoter and a weak RBS.

Final construct parts


After we conducted the necessary experiments and chose the best-performing parts, we used Golden Gate to assemble our final construct. It consists of 2 Level 2 plasmids, each of them with 4 transcriptional units. For the biofilm overexpression module we chose the P. sp. IsoF R196A mutation after we conducted statistical analysis for our c-di-GMP assays. Additionally, we selected the strongest RBS for our kill switch.
