This year, the OKstate iGEM 2024 team’s goal is to utilize synthetic biology to develop a novel treatment method targeting multi-drug resistant Staphylococcus aureus infections. We aim to accomplish this by combining two alternative antibacterial approaches. The first method is based on the concept that bacteriophages, which are viruses that targets only bacteria, can be altered to recognize specific species of bacteria, which in this case will be S. aureus. The second method is called Photodynamic Therapy (PDT) and is based on the idea that photosensitizer dyes can release antibacterial reactive oxygen species (ROS) when activated by lights at specific wavelengths.
Our basic parts collection includes the coding sequence for the E. coli DsbA signaling peptide, which is required for the translocation of any recombinant proteins across the Sec secretion machinery in E. coli. In our project, this signaling peptide is designed to be fused at the N’terminal end of our chimeric phage proteins Gp3 or Gp8. Gp3 and Gp8 are phage proteins that encode for the tail of the M13 phage. Gp3 in particular has been used for phage display studies. We have created basic parts for these two proteins. In addition, between the signaling peptide and the phage proteins, we have created three basic parts which encode for short polypeptides that are known to bind specifically to S. aureus.
