We successfully built 1 composite parts and 5 basic part for ligninolytic enzymes production in P. putida KT2004.
Our composite parts are based on the promoter BBa_5204002 and DNA of BBa_5204004. This composite part encodes a serious of proteins for maturing cytochrome C, which is composed of the basic parts of a strong promoter and transfaction sequence region, and related to manganese oxidation of P. putida.

To realize our goal to construct vector allowing P. putida KT2004 to produce multiple ligninolytic enzymes, we used 5 basic parts. There were designed to induce over expression of multiple ligninolytic enzymes in pBBR1-MCS5 plasmid, incluing multicopper oxidase (MCO), manganese peroxidases (MnP) and manganese catalase (MnC). The enzymes will meanwhile stimulate the Mn2+ oxidation reaction to produce manganese oxide, which can be served as MnP mimics with lignocellulose degradation ability. All of them are new ones that we have submitted.
