The TadA-CDd-V106W protein belongs to the CBE protein family. It was designed by members of David Liu's lab, who engineered fusions of CRISPR/Cas9 and a cytidine deaminase enzyme that retain the ability to be programmed with a guide RNA, do not induce dsDNA breaks, and mediate the direct conversion of cytidine to uridine, thereby effecting a C→T (or G→A) substitution.

Though as a Cytosine Base Editor,in addition to the risk of off-target DNA binding caused by traditional Cas9's non-specific interactions, the CBE system also has off-target risks that are independent of Cas9.But CBE what we use is evoluted from ABE by PANCE.So it is small, efficient and with low off-target activity which is totally different from simple CBE.And building upon the work from Liu's lab, we optimized the protein sequence, further reducing its off-target risk.
