One difficulty we encountered during our project was the lack of documentation on proper techniques to cultivate cyanobacteria. Thus, we aim to contribute to other iGEM teams, who want to work with cyanobacteria, by providing our own manual with all of the insights we gathered from conducting our own experiments and discussing with cyanobacteria experts. This manual includes ideal growing conditions, how to evaluate health, and transformation protocol. While we present our methods using two main cyanobacteria strains, Synechocystis sp. PCC 6803 and Synechococcus sp. UTEX 3154, it may be presumed that these techniques can be used for other strains as well.
