We have successfully constructed sacB mutants with different toxicities. At a sucrose concentration of 0.1%, the mortality rates of sacB_S164A and sacB_E262Q were 19.33% and 41.35%, respectively. Now we can provide the iGEM community with a sacB screening gene toolkit consisting of different sacB mutations and sucrose concentrations with varying mortality rates.
We have innovatively proposed a three-plasmid screening system. A reasonable combination of on-target, off-target, and screening genes can achieve high-throughput and precise screening of CRISPR Cas nucleases. The mutation, screening, and evaluation methods for MAD7 in experimental design can be applied to the modification of almost all CRISPR Cas gene editing tools. Our project design and results may inspire the future development potential of gene editing therapy for diseases.
We innovatively consolidated all the communication in the project into a new section.
On the one hand, this section can comprehensively and intuitively display all the communication completed by the project, breaking the limitations of project communication in time and space, and getting rid of the scattered communication caused by different communication characters, communication objects, and communication time. This provides project team members with comprehensive references when summarizing the project and deploying work, avoiding duplication of work and incomplete thinking among project members.
On the other hand, for people outside the project who have never known about the project, the setting of this module is to make them more convenient and quickly understand all the communication contents of the project, so that they can have a preliminary understanding when watching other modules. It presents projects as a new way of thinking for human practice, and it runs through the entire project and can be indexed to specific project content.
In summary, Communication provides a new project narrative perspective for the iGEM community, facilitating project management and content presentation.
