We are Squirrel-CHN, and this document outlines the detailed experimental protocols for our project, designed to ensure reproducibility for future teams. Our work includes constructing engineered strains, covering all steps from sequence design to plasmid extraction and PCR amplification. We also employ techniques such as gel recovery, restriction digest, ligation, and transformation. For protein expression verification, we use fluorescence microscopy, ELISA, SDS-PAGE, and Western blotting. Additionally, we integrate artificial protein reengineering with tools like AlphaFold and molecular dynamics simulations to refine and analyze our target proteins. By providing these comprehensive procedures, we aim to make it easier for future teams to replicate and build upon our work.
