Harm of the nematode
  Plant nematode is one of the important pathogens causing crop diseases in China. It is a serious threat to China's wheat, corn, rice, sweet potato, potato, soybean, vegetables, peanuts, Chinese herbs and other food and economic crops and the safety of production of important diseases.
  For example, Soybean cyst nematode disease occurs in an area of

hm2 per year, which can cause soybean yield reduction of
5%-10%
, and the serious occurrence of plot yield reduction of more than

, or even no harvest. Losses caused by soybean cyst nematode disease amounted to more than
600 million yuan
  With global climate change, cultivation system reform and the rapid development of large-scale,mechanised and high-value agriculture, plant nematode diseases are in a serious trend of occurrence, and will rise to be the second largest plant disease in China.

  Nematodes use oral needles to secrete secretions from the oesophageal glands into host plant cells and induce host cells to form specialised feeding sites.

  The establishment and maintenance of feeding sites enable nematodes to absorb large amounts of nutrients from the plant, which promotes the growth and development of nematodes and induces disruptions in the partitioning of photosynthetics.

  Common nematode control methods, such as chemical control and land management, have problems such as pollution of the environment and poor quick-acting properties.

  To mitigate the economic impact of nematode diseases and ensure global food security, we have designed this year's project:
Bacillus Gemini: Plant Guard & Nematode Buster
. This innovative method utilizes two types of plant beneficial bacteria as engineered bacterial chassis: Bacillus subtilis (Plant Guard) and Bacillus velezensis(Nematode Buster), allowing them to collaboratively exert their effects.

1. Plant Guard
(Bacillus subtilis):
-Its primary role is defense against nematodes.
2. Nematode Buster (Bacillus velezensis): -Its primary function is to attack nematodes.
3. Enhancing Colonization Abilities: -It enables bacteria to form stronger symbiotic relationships with plants and effectively carry out their protective functions.

1. Plant Guard (Bacillus subtilis):
-Bacillus subtilis is widely distributed from the root zone to the leaf surface of plants.
-Its primary role is defense against nematodes.
-It expresses virus-like particle vectors containing RNA for targeted plant immune regulation.
-Also,VLP carries externally linked plant immune-activating proteins, contributing to plant immune induction.

2. Nematode Buster (Bacillus velezensis):
-Bacillus velezensis predominantly resides in plant roots and can form biofilms
-lts primary function is to attack nematodes.
-Upon sensing hormonal changesin response to nematode damage(salicylic acid), the engineered bacteria synthesize virus-like particles.
-These particles carry anti-nematode protein and include antisense RNA targeting key genes in the life cycle of nematodes.
-Furthermore, Bacillus velezensis secretes a novel anti-nematode molecule called trans-Aconitic acid.
-The engineered bacteria exert a comprehensive lethal effect on nematodes through the aforementioned series of mechanisms.

3. Enhancing Colonization Abilities:
We enhance the colonization abilities of the engineered bacteria.
By enhancing the expression of cell wall phosphates and surface starch-like proteins,
both strains exhibit better survival and adhesion capabilities.
This enables them to form stronger symbiotic relationships with plants and effectively carry out their protective functions.

  In summary, the use of these Bacillus as biological control agents holds promise as a durable strategy against plant-parasitic nematodes in agriculture. Their multifaceted mechanisms, including direct antagonism, adhesion protection, and immune system activation, contribute to reducing nematode damage and enhancing plant defenses.
