Introduction

The goal of our team was to design a modular gliadin-degrading enzyme secretion system that could be transformed into GI tract bacteria. To do this, our team split into 4 main research areas:

  1. Constructing the backbone of for our modular system and testing transformation methods with our backbone.
  2. Constructing promoter and fluorescence protein systems with our backbone to characterize the strength of various promoters and promoter regions.
  3. Constructing and testing induction systems and characterizing signal peptides in our specific environment.
  4. Constructing and expressing gliadin-degrading enzymes constructs in GI tract bacteria, and isolating LABs from fermented foods.

During the experimental project, we used the protocols below to conduct our various experiments across the 4 main goals. Additionally, we constantly refined our experimental design and approach based on feedback from our results, literature, mentors, and our stakeholders.
