Introduction
The goal of our team was to design a modular gliadin-degrading enzyme secretion system that could be transformed into GI tract bacteria. To do this, our team split into 4 main research areas:
- Constructing the backbone of for our modular system and testing transformation methods with our backbone.
- Constructing promoter and fluorescence protein systems with our backbone to characterize the strength of various promoters and promoter regions.
- Constructing and testing induction systems and characterizing signal peptides in our specific environment.
- Constructing and expressing gliadin-degrading enzymes constructs in GI tract bacteria, and isolating LABs from fermented foods.
During the experimental project, we used the protocols below to conduct our various experiments across the 4 main goals. Additionally, we constantly refined our experimental design and approach based on feedback from our results, literature, mentors, and our stakeholders.
